Yakuman DS features a total of 21 characters, 6 of which are playable from the start, while 7 are unlockable (the remaining 8 are only featured as opponents).
In detail the starting characters are:

- Princess Peach
- Donkey Kong
The unlockable ones are:
- Wario
- Waluigi
- Bowser
- Princess Daisy
- Petey Piranha
- Bowser Jr.
- Toadsworth (secret)
While the ones only featured as opponents are:
- Goomba
- Shy Guy
- Koopa Troopa
- Boo
- Birdo
- Diddy Kong
- Hammer Bro. (unlockable challenge)
- Toadette
The game also features supporting characters such as Lakitus and Bob-Ombs.
